چکیده

Natural Bile Pigments, Coprobiliverdin III, Model Chromophore, Spectroscopy, MO-Calculations The synthesis of l5N-labeled coprobiliverdin III, a bile pigment with four conjugated pyrrole nuclei and four carboxylic acid side chains, is described. Coprobiliverdin is structurally characterized by chemical and various spectroscopic methods (e.g. chromic acid degradation, UV-vis-, IR-, mass spectroscopy). The results of 15N NM R studies reveal the potential value of this compound as a probe for structural interactions in biliproteins and as a reference for the future investigation of natural bile pigments deriving

Structure elucidation of azotobactin 87, isolated from Azotobacter vinelandii ATCC 12837.

Chromopeptide siderophores (azotobactin 87-I and -II) were isolated from an iron deficient culture medium of Azotobacter vinelandii ATCC 12837 (= DSM 87). Their structures were elucidated by chemical degradation studies and spectroscopic methods, especially 2D-NMR-techniques. Total assignments of 1H-, 13C-, and 15N-resonances based on 2D-HOHAHA-, 1H/13C-HMQC-, 1H/13C-HMBC-, 1H/15N-HMQC/TOCSY-, ...

Using 15N Dilution Method to Correct for Microbial Contamination When Assessing in situ Protein Degradability of Fresh Ryegrass

This experiment was done to investigate microbial contamination and in situ disappearance rates of dry matter (DM), N and 15N of fresh labeled ryegrass. Perennial ryegrass (Lolium perenne) were labeled with 15N during growth in a glasshouse, harvested at 4th leaves stage and were incubated up to 34 h in situ in the rumen of 3 individually housed sheep. The animals were fed 800 g/d chopped alfal...

The site of attachment of retinal in bacteriorhodopsin. A resonance Raman study.

The retinal chromophore of bacteriorhodopsin is attached as a Schiff's base with the epsilon-amino group of a lysine residue. The site of attachment has now been investigated by the use of resonance Raman spectroscopy which has previously been shown to be sensitive to 15N isotope substitution at the Schiff's base. Bacteriorhodopsin samples obtained from bacteria grown in a medium containing eit...

Chromophore-in-Protein Modeling of the Structures and Resonance Raman Spectra for Type 1 Copper Proteins

Geometries and resonance Raman (RR) spectra have been modeled for the type 1 Cu active sites of plastocyanin and azurin, as well as two azurin site-directed mutants, M121G and H46D. Using force constants for the Cu coordination group chosen to fit the RR spectra in conjunction with the AMBER force field, we calculated geometries and vibrational spectra. The fitting procedure utilized a chromoph...

Light-induced chromophore activity and signal transduction in phytochromes observed by 13C and 15N magic-angle spinning NMR.

Both thermally stable states of phytochrome, Pr and Pfr, have been studied by (13)C and (15)N cross-polarization (CP) magic-angle spinning (MAS) NMR using cyanobacterial (Cph1) and plant (phyA) phytochrome sensory modules containing uniformly (13)C- and (15)N-labeled bilin chromophores. Two-dimensional homo- and heteronuclear experiments allowed most of the (13)C chemical shifts to be assigned ...
